Identification of specific DNA or RNA sequences in these situations could inform medical professionals and potentially curb health crises. These reactions can even be run without electricity, which is particularly attractive for point of care and limited resource molecular diagnostics. Isothermal DNA amplification reactions that require less energy, time and equipment than PCR have become an increasingly popular alternative for molecular detection. While polymerase chain reaction (PCR) provides a gold standard for accuracy and sensitivity of nucleic acid detection, it requires complex laboratory equipment and trained personnel. The reaction mechanisms are (i) positively cooperative multistep binding spurred by two trigger binding sites on the template (ii) gradual template deactivation (iii) recycling of deactivated templates into active templates and (iv) polymerase sequestration.
